Shiseido Develops a New Introductory Ingredient That Dramatically Enhances Skin Penetration of Drugs

Shiseido has developed the world's first infusion-promoting ingredient that dramatically increases the penetration rate of drugs while also ensuring skin safety by applying the phenomenon in which normally solid substances interact with specific molecules to liquefy. developed.

In order to quickly penetrate the drug into the target site of action in the skin, it was necessary to lower the skin barrier function, and there was an issue with skin safety. By utilizing it, it became possible to efficiently penetrate the drug while maintaining safety.

With the newly developed ingredients to promote introduction, we aim to create highly functional and effective skin care products with excellent permeability, as well as high quality that pursues gentleness on the skin.

In recent years, there has been a growing demand among consumers for the extremely high effectiveness of quasi-drugs. In order to realize this, the company is promoting technological innovation in the "drug development area" based on "development of original ingredients" and "research on active ingredients" to quickly deliver active ingredients to target areas deep in the skin. We are working hard to deliver more.

On the skin surface, there is a layer that has a high affinity for oil and has the property of repelling water. On the other hand, many of the agents used in cosmetics are designed to have a high affinity with the base water, so they repel water. Due to its properties, it is difficult to penetrate the skin, and drug introduction accelerators have been developed for some time.

While the introduction enhancer promotes the penetration of the drug, it has a surfactant-like structure and properties, so it penetrates into the stratum corneum, lowers the lipid barrier function, and penetrates deep into the skin, damaging cells. Issues such as giving

Therefore, this time, we tried to develop an unprecedented new introduction accelerator in order to deliver the active ingredient to the site of action without fail and to provide a product that is both highly safe.

In developing a new introduction accelerator, we focused on a new type of liquid called an "ionic liquid," which becomes liquid at a temperature lower than the melting point of the original material by combining substances with high melting points.

Alkylbetaine (AB), which is commonly used as an introduction accelerator, was combined with various substances, and more than 100 combinations of liquefied combinations and their blending ratios were verified, and alkylbetaine (AB) and xylitol ( XY) combination (AB/XY complex) is liquefied and has an excellent permeation promoting effect into the skin.

In addition, when we evaluated the penetration enhancement effect of 4MSK (4-methoxysalicylic acid potassium salt), a whitening agent, using human skin, we found that the penetration rate of the new introduction-enhancing ingredient was higher than that of the drug alone. was increased by about 3.7 times, and the amount of penetration was increased by about 3.5 times.

Common uptake-enhancing agents infiltrate into the lipids of the stratum corneum, disrupting the ordered lipid structure and promoting the penetration of coexisting drugs. It showed a penetration promotion mechanism that creates a gap and creates a permeation route for the drug, leading to excellent effects.

Next, we considered not only improving the penetration promotion effect of the drug, but also improving the safety of the skin. Xylitol covers part of the structure of substances that are generally thought to damage skin cells, thereby mitigating damage to cells. The stimulating component AB/XY complex was exposed to living skin cells and cell viability was observed.

Despite the fact that the experiment was conducted under experimental conditions in which it was difficult for cells to survive with the transduction promoter alone, the number of cells that lost mitochondrial activity even after exposure to the new transduction promoter AB/XY complex complex was small. As with exposure to , no significant reduction was observed.

In the future, we aim to provide beauty solutions that exceed customer expectations by combining new introduction-promoting ingredients that apply the phenomenon of "liquidizing at room temperature" and various drugs to further improve the effectiveness of quasi-drugs.

In addition, for the development of other functional ingredients, we will continue research that utilizes the phenomenon that "usually solid substances interact with specific molecules to liquefy." We will continue to innovate.
